چکیده مقاله:

Background & Objectives: Pseudomonas aeruginosa is a common cause of nosocomial infection. OprD protein is a specific protein regulating the uptake of carbapenem antibiotic. Loss of OprD is the main mechanism of Pseudomonas Aeruginosa resistance to carbapenem. In this study, the presence of OprD gene is investigated in isolated Pseudomonas Aeruginosa in burn patients of Ghotboddin hospital in Shiraz. Material & Methods: 66 Pseudomonas Aeruginosa were isolated from wound specimens of 250 burn patients. Strain characteristics were confirmed by biochemical tests. Antibiogram was done via disc diffusion method. Finally, OprD gene was investigated by PCR. Results: Isolated Pseudomonas Aeruginosa showed more sensitivity to chloramphenicol and colicitin and more resistance  to ciprofloxacin, gentamycin, cefotaxim, ceftazidin, imipenem, meropenem, and erythromycin. 61 percent of isolates were positive for OprD gene by PCR. Conclusion: The findings of this study revealed that Colicitin and chloramphenicol are more effective in treatment of Pseudomonas Aeruginosa infections in burn patients, and deletion and mutation in OprD gene cause bacterium resistance to carbapenem antibiotic.

Molecular Investigation of Outer Membrane Channel Genes Among Multidrug Resistance Clinical Pseudomonas Aeruginosa Isolates

Background: Multidrug resistance Pseudomonas aeruginosa (MDRPA) is most important issue in healthcare setting. It can secrete many virulence effector proteins via its secretion system type (T1SS-T6SS). They are using them as conductor for delivering the effector proteins outside to begins harmful effect on host cell increasing pathogenicity, competition against other microorganism and nutrient ...

Role of PA3574 (nalD) gene in development of ciprofloxacin resistance in Pseudomonas aeruginosa isolates

Background & Aims: Pseudomonas aeruginosa is a gram-negative opportunistic pathogen and one of the mortality causes of nosocomial infections. One of the drug resistance mechanisms in P.aeruginosa is mutation in negative regulator genes of mexAB-oprM efflux pump system such as nalD. The aim of this study was to investigate the role of nalD mutations in P. aeruginosa isolates of Guilan province i...

Antibiotic resistance in clinical isolates of Pseudomonas aeruginosa in Jamaica.

OBJECTIVE To assess antibiotic resistance in clinical isolates of Pseudomonas aeruginosa in Jamaica, and to obtain baseline information on the presence of this important pathogen. METHODS A total of 51 isolates of Pseudomonas aeruginosa, obtained from 162 clinical specimens from major hospitals and laboratories in seven parishes in Jamaica, were analyzed between May and August 2002. Isolates ...

Antibiotic resistance profiles of Pseudomonas aeruginosa isolates containing virulence genes

Background: A most common opportunistic pathogen, Pseudomonas aeruginosa is present in both humans and animals and responsible for various nosocomial infections and healthcare settings related infections. Different virulence genes like; oprL (membrane lipoprotein L) and toxA (exotoxin A i.e. ETA) in P. aeruginosa, assist in its pathogenicity, toxicity and contribute to high antibiotic resistanc...

Contribution of outer membrane efflux protein OprM to antibiotic resistance in Pseudomonas aeruginosa independent of MexAB.

A Pseudomonas aeruginosa strain carrying an insertion of an omega Hg interposon in the mexB gene (mexB::omega Hg; strain K879) produced markedly reduced but still detectable levels of OprM, the product of the third gene of the mexAB-oprM multidrug efflux operon. By using a lacZ transcriptional fusion vector, promoter activity likely responsible for OprM expression in the mexB::omega Hg mutant w...

Detection of outer membrane porin protein, an imipenem influx channel, in Pseudomonas aeruginosa clinical isolates.

Decreased permeability to imipenem is the most frequent mechanism of imipenem resistance in Pseudomonas aeruginosa. We have determined the presence of OprD porin protein, an imipenem influx channel, in 70 carbapenem-resistant P. aeruginosa clinical isolates by Western blot analysis using rabbit anti-OprD polyclonal antibody. Ninety-eight percent (54 of 55 isolates) of imipenem-and meropenem-res...
